Mirren, 76, was seen comforting Gaga and wiping tears from her eyes as the Grammy Award-winning singer cried in her arms.
On Sunday, Gaga was nominated for Best Actress in a Leading Role for her portrayal of Patrizia Reggiani in the Ridley Scott-directed murder drama House of Gucci.
In the meantime, Mirren received the SAG Life Achievement Award. With a total of 13 SAG Award nominations and five SAG Award wins, she is the most decorated SAG Life Achievement honoree to date.
"Lifetime achievement," she said in her acceptance speech, "sounds so grand; I suppose I'm still awake, so by that measure, I'm eligible." She went on to say that her "mantra" is "Be on time and don't be an ass" and that it is the driving force behind all of her accomplishments thus far.
"Thank you, S-A-G, so much for this," Mirren continued as the audience laughed. "I hate to state SAG at my age, but it's always S-A-G for me."
"I am immensely proud while also knowing that I do not deserve it, and therein lies the conflict that I believe you all recognize: insecurity versus ego. That cocktail, I believe, is what most actors drink in the evening as they reflect on the gift they received when they decided to pursue a career in acting, "She continued.
"I've been angsting regarding what to say tonight," Mirren said, "and then it came to me: to all those individuals with whom I've shared my professional life, the actors!" Mirren also mentioned her battle with knowing what to say during this speech.
Although Gaga did not receive an award on Sunday, she appeared to have a great time catching up with her celebrity friends at the event, including her A Star Is Born co-star Bradley Cooper.
The couple smiled as they reunited more than three years after their award-winning film was released, lovingly embracing one another while attending the star-studded event.
During her acceptance speech, Ariana DeBose gave a sweet shoutout to the "Rain On Me" singer for an excellent performance by a female actor in a supporting role. "Thank you all, all of you staring at me, so many of you, dear God," DeBose said after winning for her role as Anita in West Side Story. "Hello, Lady Gaga! You're incredible!"
